Item Formulation of Nano-sponge for Diabetes treatment(Library Information Services COMSATS University Islamabad Lahore Campus, 2023-02-27) Sana; SP22-R06-031; Dr. Arsalan Ahmed Ansari; LHR TP 8667In Diabetes research, the advancement of nanotechnology has made it easier to characterize nanosponges by having an ability to absorb the glucose at the elevated level from the blood by a novel method. It could significantly enhance the diabetic's quality of life. Our focus was on recent developments in diabetes filed research at the point of intersection with nanotechnology. Recent progress in the field of diabetes research at its interface with nanotechnology was our focus. The goal of this work is to improve therapeutic bioavailability, stability, and biodegradability. For this purpose, the herbal material as Basil seeds were targeted due to its good potential for diabetic individuals and its mucilage contains glucomannan which is a polysaccharide. The mucilage of basil seeds was used for the formulation of the nanosponge. The physical and chemical characterizations of the glucomannan (GM) and glucomannan Nanoparticles (GMNPs) were performed by UV, FTIR, XRD and their particles size and charge were determined by using DLS and Zeta Analyzer. The morphological study was performed by the FESEM, and the distribution of particle size was determined by histogram the particle size at 500nm centered around the 25nm. The formulated Nanoparticles were also evaluated by their Solubility analysis and the Stability of the Nanoparticles in different fluids as SBF (Simulated Body fluid) having pH=7.4, SGF (Simulated Gastric Fluid) having pH=1.2, SIF (Simulated Intestinal Fluid) having pH=6.8, and water. Our nanoparticles showed good solubility in the fluid (SGF) at the pH=1.2 which is acidic, as well as observed good stability behavior till 24 hours and then start to degrade, which confirms that nanoparticle will easily move into the intestine without degradation and agglomeration. In the SIF, the nanoparticles showed stability till 38 hours which confirms that nanoparticles will absorb more glucose in intestinal fluid as most of the digestion takes place there. And in case of simulated body fluid, nanoparticles showed less solubility in the SBF due to the agglomeration of nanoparticles. An efficient study as sugar intake study was also conducted to check the glucose absorption in these different fluids and in the combination with the interfering specie as fructose, xi the overall trend with respect to the time as 3hours, 5hours,7hours, the maximum absorption of sugar was increasing. In the first 3 hours the nanoparticles shown the maximum absorption. Studies conducted in vitro assess the safety and effectiveness, compatibility of these nano systems in the treatment of diabetes. The cell viability of the glucomannan and glucomannan nanoparticles were also evaluated by using Mouse fibroblast NIH3T3 in Dulbecco’s Modified Eagle Medium (DMEM) by using Alamar Blue Assay in three days study by using different concentrations of nanoparticles as in 100μl, 50μl and 25μl. In the case of glucomannan, the percentage of cell viability was observed 87.82% in 100μl and in case of glucomannan nanoparticles shown the percentage of cell viability 93.92% which confirmed that our nanoparticles are non-toxic, biocompatible.
