    Development of Drug Loaded Injectable Bioactive Glass for Dental Applications

    Conventional therapies practiced for the treatment of dental infections are limited and invasive. Thus, in recent years, injectable hydrogel systems have emerged as a promising approach for tissue repair and localized drug delivery. These systems offer advantages such as ease of administration, minimal invasiveness, faster recovery, and long-term efficacy. That is why, in this study, we synthesized a novel thermoresponsive, injectable hydrogel using biocompatible and biodegradable components, namely chitosan and alginate, in combination with the bioactive glass that will be used to promote bone regeneration along with the antibacterial properties. Various concentrations of the primary polymers and selenium-substituted bioactive glasses were investigated, and the hydrogels were characterized using techniques such as X-ray diffraction (XRD), Fourier-transform infrared spectroscopy (FTIR), and contact angle measurements. Gelation behavior and setting time of the hydrogels were analyzed both with and without the presence of bioactive glass by employing tube inversion method and a setting time of 13 minutes was achieved for the prepared hydrogel system. Their injectability was assessed using syringe needles of different gauges like 17G, 18G, 19G and was found to be in accordance with their applicability as minimally invasive procedures. Furthermore, antibacterial studies were conducted against S. mutans, revealing significant antibacterial activity of the drug loaded hydrogels containing selenium-substituted bioactive glass with the maximum inhibition shown by the hydrogels with highest percentage of selenium.

    Hydrogels with Antibacterial Properties for Potential Treatment of Periodontitis

    Periodontitis is a severe inflammation of specialized tissues that support and surround the teeth called periodontium. Periodontal tissues weaken over the passage of time due to the persistent bacterial attack and are difficult to recover once they are lost and damaged. For tissue repair or wound healing, it is important to develop a biomaterial which would have such qualities that make it most suitable for the regeneration of bone tissues along with antibacterial properties. Bioactive glasses (BG) have been studied for their biocompatibility and ability of bone regeneration and hydroxyapatite formation in the body. Moreover, Acetanilide (Drug) has been found to possess analgesic, anti-pyretic as well as anti-bacterial properties and was expected to be better choice for developing a hydrogel in combination with bioactive glass that heals or repair the bone tissue. In this study a thermo-sensitive, anti-bacterial and biodegradable chitosan-based hydrogel was developed by Sol-Gel method using different concentrations of bioactive glass in combination with chitosan and acetanilide. Physiochemical properties of hydrogel were studied through various analytical techniques such as FTIR, XRD and contact angle. Gelation and setting time of bioactive glass based hydrogels were studied and it was observed that gelation and setting time were affected by the different concentrations of bioactive glass. The injectability and flowabililty of hydrogel was also investigated using needles of different gauges. Anti-bacterial potential was analyzed against two bacterial strains, E.coli and S.aureus, in which chitosan based hydrogel with bioactive glass and Acetanilide (CS-BG-Aa) showed effective anti-bacterial activity.

    Synthesis and Characterization of Injectable, Biocompatible and Biodegradable Hydrogels for Biomedical Application

    Periodontitis is a common oral infection that affects about 10-15% of global population. Periodontitis is characterized by gum inflammation, gingival recession, tissue loss, and dental pocket formation. Different approaches like tooth scaling, ultrasonic scalars, lasers, and root planning as well as surgical procedures have been employed but these mechanical treatments are limited and invasive. For tissue repair or wound healing, it is important to develop biomaterials which have ability of regeneration of bone tissues along with antibacterial properties and would be less invasive in nature. Injectable hydrogels are crucial in this area because they have mechanical and structural characteristics comparable to many body tissues and the artificial extracellular matrix (ECM) and can be delivered to the body in the least invasive manner. In this study a novel thermosensitive, injectable, biocompatible, and biodegradable Chitosan-Alginate based hydrogel was synthesized in combination with bioactive glasses. Different concentrations of primary polymers and copper substituted bioactive glasses were studied and characterized by employing various analytical techniques including XRD, FTIR, Contact Angle, Zetasizer and Inverted microscope. Gelation effect and setting time of these hydrogels were analyzed with and without Bioactive glass. The injectability of these biomaterials was tested via syringe needles of various gauges. Bioactivity and degradability studies were performed along with antibacterial applications against E. coli and S. aureus in which the hydrogels with copper substituted bioactive glass demonstrated positive anti-bacterial activity.

    Synthesis and Characterization of Metal Substituted Bioactive Glass for Treatment of Periodontal Disease

    Periodontitis is a serious gum infection that damages soft tissue and if not treated well can destroy bone resulting in tooth loss and is caused by different bacteria such as Aggregatibacter actinomycetemcomitans, Porphyromonas gingivalis and Tannerella forsythia. Roughly 50% world population is affected with it and 750 million people suffer from its severe form. The damaged bone can be regenerated in different ways, including using one’s own bone, or using donor’s bone and third way by using different synthetic bone fillers including bioactive glasses. Bioactive glasses (BGs) are non-crystalline biomaterials that are able to bond soft and hard living tissues and dissolve over time to stimulate growth of new tissues. In this research, an effective approach of periodontal treatment is to treat it with nano-sized bioactive glass prepared by co-precipitation method in which metal copper and fluoride ions are doped to increase its antimicrobial properties as well as strength. Different concentrations of copper-fluoride (Cu-F) doped bioactive glass have been prepared and characterized by different analytical techniques including FTIR, XRD and SEM-EDX to confirm the successful incorporation of copper and fluoride ions. The results showed that metal doped bioactive glasses were successfully prepared.

    Synthesis of Biomolecule Loaded Bioactive Glass and its Antibacterial Properties

    Bioactive glasses (BGs) are synthetic bone fillers that have ability to replace damaged bone, facilitate tissue regeneration and integrate with body environment. In this study we used bioactive glass prepared by co-precipitation method and used doxycycline as a biomolecule. Doxycycline (DOX) was loaded on bioactive glass with different concentrations (2mg/mL, 4mg/mL and 8mg/mL). Both, simple BG and BG/DOX was characterized using Fourier Transform Infrared spectrophotometer (FTIR), X-Ray Diffraction (XRD), and Scanning Electron Microscopy (SEM). Antibacterial studies were conducted against E. Coli and S. Aureus bacteria. The results showed that BG/DOX was affective against these bacterial strains as they inhibited their growth. The zone of inhibition was 19.5mm, 26mm, 26.5mm and 22.5mm, 28mm, 24mm for BG/DOX-2, BG/DOX-4 and BG-DOX-8 against E. coli and S. aureus respectively. There was not much difference in zone of inhibition by changing concentration from 4mg to 8mg. Hence, 4mg is considered as an optimum concentration. Therefore, Doxycycline drug loaded BGs can be used to enhance the antibacterial properties of BGs, in tissue engineering applications, and in root implants of teeth's for long term effect.
